Can I change a comment in the General Ledger in Sage 100 V5.00.8.0?

I made a mistake in the description in the General Ledger. Can I change the comment or am I stuck with it?

Assuming you are talking about the comments on a general journal entry, short of using data file display and maintenance (which I really don't advise), you are pretty much stuck with it.
