running the sage 300 CRM Setup, have a following error Please verify that the server name and TCP/IP port number specified are correct'.

If you are running SQL server express then you need to enable the TCPIP protocol. If you have a firewall running then you need to open port 1433 for SQL server.

currently using SQL Server 2014 not SQL server Express, the firewall was turned off. How do I open port 1433?

You don't have to open port 1433 if the firewall is off. Double check your work, either the SQL server name is incorrect or the firewall is running and port 1433 is not open.

The firewall is turned off but we are currently using E-Scan anti-Virus which might have its own firewall, will that be the case?

Disable all firewalls or open port 1433 in all firewalls.
